In Train-Sim an "activity" is like an adventure in Flightsim. It's a preset trip that you take with a train and can be simple (just driving station to station) or complex (switching, meets with other trains, following a schedule). It would appear that you can set up just about anything that a real railroad would do. Sometimes special challenges are included, like one of the steam train trips only has limited water available so you cannot just drive the route flat out, you also need to manage the boiler to use as little water as possible.
It's also where you make up consists (a consist is the locomotive(s) and car(s) that make up your train), though for some reason they've made this a bit hard to get to.

Robert, what do you think is the chance of getting Microsoft or or Kuju to do a write up better docs for the editor for this game? Nothing official but maybe something more detailed then what we have now. If we tell them that it will help fans of the game to make more routes, trains, and activites. That of course will make the game even more popular and sales will even grow larger.

Well, there's a book coming out in a few weeks. And, I'm sure that if you hang out here you'll see more and more info posted about how to do various things.
